Maintaining Signaling Balance
Because PI3K–AKT regulates fundamental cellular processes, its activity must be carefully controlled. Cells therefore rely on regulatory mechanisms that determine the intensity and duration of the signal.
One of the most important negative regulators is PTEN, a phosphatase that counteracts PI3K activity by reducing PIP3-dependent signaling. The balance between PI3K-mediated PIP3 production and PTEN-mediated signal attenuation contributes to the precise control of AKT activation.
This regulatory balance illustrates an important principle of cellular signaling: biological responses depend not only on whether a pathway is activated, but also on how strongly, how long, and in which cellular context it is activated.
